High-performance Liquid Chromatography
Background
In high-performance liquid chromatography (HPLC), a mixture sample is being pumped at a high pressure to pass through a column (stationary phase) and its components flow out from the column at different time points to be analyzed by the connected detectors. Its mechanism resembles that of fast protein liquid chromatography (FPLC), but it has a higher separation resolution because of the higher pressure and the smaller resin particles of the columns. According to the purpose of HPLC, it can be divided to be analytical HPLC and preparative HPLC. They differ in column volume, column particle size, HPLC device pump capacity, and HPLC device tubing size. Analytical columns typically have particle sizes of 3-5 µm whilst preparative column has particle sizes of over 10 µm.
