Protein Nanocage Mass Spectrometry

Mass spectrometry (MS) is a powerful and highly accurate technique to identify proteins and analyze protein primary sequence. In the application of ferritin and virus-like particles (VLPs) as drug delivery platforms and vaccines, correct molecular weight and a 100 % sequence accuracy of the protein nanocage are indispensable for the achievement of the theoretical conformation and bioactivity. In many cases, mutations or fusions with foreign peptides of protein nanocages are employed to incorporate desirable functions with protein nanocages. The verification of these intended alterations is necessitated before any further characterizations. When using eukaryotic expression hosts to express protein nanocages, the post-translational modification sites and types need to be identified and their impact on protein nanocage function needs to be assessed. In addition, during the preparation of protein nanocages, unexpected degradations of protein nanocages are likely to occur and cause problems in the sequential characterizations and evaluations. Mass spectrometry is a direct and highly sensitive tool to shed light on the mechanism behind these degradations.
